The aqueous layer will typically not turn black when NaOH is combined with a relatively pure DMT salt solution; I have only observed this blackening when basing root bark. The water should go from a translucent/transparent clarity to opaque after the base is added.
I would do 3-4 pulls with 75-100ml of solvent each. Roll the solvent/aqueous mixture on the ground in a sealed cylindrical container (canning jar) to mix the layers. I usually get pretty bad emulsions if I shake or vigorously stir.
I have had good results when heating in a water bath but I have never made a comparison with an unheated extraction.
Adding a significant amount of pure table salt to the water can help drive the alkaloids into the naphtha.
