Chemical Composition of Traditional and Analog Ayahuasca

The study aimed to determine concentrations of the main bio- and psychoactive components of ayahuasca used in different locations and traditions. We collected 102 samples of brews from ayahuasca-using communities. Concentrations of N,N-dimethyltryptamine (DMT), tetrahydroharmine, harmine, and harmaline were determined by ultra-high performance liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS). Qualitative analyses for non-traditional additives (moclobemide, psilocin, yuremamine) were performed by high resolution mass spectrometry. Higher and more variable concentrations of DMT in neoshamanic ayahuasca samples compared to indigenous samples may indicate use of higher and more variable proportions of DMT-containing admixture plants. From European samples, we found two related samples of analog ayahuasca containing moclobemide, psilocin, DMT, yuremamine, and very low concentrations of B. caapi alkaloids. Some analogs of ayahuasca (Peganum harmala, Mimosa tenuiflora) were used in Europe. No analogs were found from Brazil or Santo Daime ceremonies in Europe.

It doesn't quite fit the hypothesis that harmaline gets reduced to THH in the decoction process...?

As seen in Figure 3, there was also
a trend toward higher DMT/HME ratios among Daime

Interesting find Jagube & thanks for link to paper ShamensStamen. It is the Santo Daime vegetal group that consistently makes brews with next to zero levels of harmaline, saw this in a study. Perhaps we should study how they brew, and if they use vitamin c? and for how long? Vitamin c is after all just like Dr. Shulgin said "an efficient hydrogen donor."
 
A little more from the study: You should know if your harmaline gets converted to tetrahyroharmine, as when mixed with water and smeared on a paper plate, it will glow blue (THH) instead of green (harmaline).

In TIHKAL, 300mg of tetrahydroharmine (THH) is equated by one psychonaut to the closed eye visionary (CEV) power of 100mg harmaline, but without all the nausea and dizziness. It glows blue when mixed with water under blacklight, like LSD or psilocin & has a metallic-like lingering taste with a 10.5 hour half-life.

If we could figure out if/how brewing methods have an impact on this, I would be thrilled to make a big section about it in the Aya sticky (I'm in the middle of reworking it).

Is it possible that the differences are simply a variation in alkaloids between types of vine?

I would be all in to brew/bioassay/take pretty pictures under blacklight for the team. :thumb_up:

Adding vitamin C to a brew can get disgusting quickly, so I'd be curious to hear if anyone knows anything about minimum effective amounts?

Great point ms_manic_minxx.:) Professor8 here at this forum back in 2010 made THH from harmaline using vitamin C, a kind search will pull up his procedure: "Conversion of Harmine->Harmaline->THH by simple boiling." I agree with his statement. Should add that music sounds quite incredible on a combo of 150mg or more of THH + 60mg sublingual "under the tongue" HPBCD DMT held under tongue for 15 minutes as tetrahydroharmine imho breaks down the filters or barriers in the mind, so that "mind at large" can be let loose, very similar to listening to music on cactus.
