Yesterday I had my first experience with a sonicator. I have a small 50-watt one, used for jewelry, cleaning small instruments, etc. Due to its low power, it was used after a 10X freeze/thaw cycle to optimize lysis. I'll finish the entire extraction today, so I can't comment on the results yet. But at first glance, I notice that the unsonicated MHRB powder is coarser than the already sonicated powder. This gives me hope that my device is indeed useful. I sonicated 50 grams of powder, divided into five parts of 10 grams each, for 60 minutes (eight times of 7.5 minutes, with breaks between each sonication period), stirring constantly and slowly to prevent the powder from settling at the bottom of my sonicator. A 200-watt sonicator would do it in minutes, but I don't have that kind of power. It was many hours of sonication!
The entire process was carried out with 250 ml of distilled water with a pH of 5.5 (acidified with a bit of vinegar) and 1.3 grams of NaCl dissolved in the water, previously heated to 60° Celsius / 140° Fahrenheit. The liquid was always reused with each of the five batches of powder.
