Tincture from 5x leaf?

Migrated topic.

Hi there!

I have 1g of '5x' leaf that I have started experimenting with again. Smoking it hasn't produced the effects I had been hoping for, even when smoking 100mg+ of it, which is supposedly above the threshold for at least some breakthrough effects. I was wondering if I could soak it in ISO to extract the goodies, evap and then dissolve in some ? to make a tincture for sublingual application. I would rather administer it this way as I am trying not to smoke anything at the moment.

Sorry if I have missed a tek outlining how to make a tincture, I don't have access to my usual internet facilities at the moment. I guess my main question is can an extract be easily re-extracted, and how would I go about making a tincture?

Many thanks in advance,

P x

I've wondered about that too. In the thread VoidTraveler shared,

It seems like not only salvinorin A is responsible for the effects to work sublingually.

and that brings us to the description of the tincture commercialized by Daniel Siebert, where it states

This ultra-concentrated tincture of Salvia divinorum has been carefully prepared using a unique new separation process developed by Daniel Siebert. This innovative process selectively extracts and concentrates a group of natural leaf components that enhance the solubility and absorption of the active principal, salvinorin A.

It would be interesting to expand this point. If the salvinorin in your x5 extract has been infused into salvia leaf, one would expect these "natural leaf components" to be present as well, only in 1/5 proportion. The fact this allegedly decreases the efficiency of tincture is quite curious, I'll look into it when I have time.

Yes, tinctures are most effective when they include some plant "impurities" rather than pure salvinorin A. There are basically two possible reasons for this:

1.) The compounds facilitate the absorption of salvinorin A through the oral mucosa.

2.) The compounds promote the formation of emulsions. Salvinorin A is fairly soluble in ethanol, but the solubility drops rapidly as the concentration of water increases and the concentration of ethanol decreases. As the tincture is diluted by your saliva, salvinorin A is apt to drop of solution (and when it drops out of solution, bioavailability is extremely poor). But if the plant compounds promote the formation of emulsions, they help to maintain salvinorin A in a more bioavailable form (as a component in an emulsion rather than a precipitate).

I suspect the second explanation is correct.

As to preparing a tincture from 5x extract: you'll have better luck with acetone than i-propanol. Sphere put together some good pdfs on the subject (see, for example, Salvia Divinorum Salvinorin Extraction and Refinement FAQ)

Another point to bear in mind: Extract-enhanced leaf products rarely contain the concentration of salvinorin A that they ought to. Your 5x might be no more potent than plain leaf. Bottom line: dosing extracts is even more of a crapshoot than dosing plain leaf (which itself can vary pretty widely in salvinorin A content).

I would use very cold acetone strait from freezer and soak all of it in it for about 3 minutes. No need too much acetone just to cover it all. Then repeat it another 2 times and evaporate it all on around 250 - 500mg of Salvia or any leafs. Use less plant material if you want it to be very strong.

After you done be careful with dosage or you get your ars kicked like me 😁

Just for a quick reference.

Salvinorin A relative solubilities :

Acetone
23 mg/mL (43.5 mL/g)
Methyl Alcohol (Methanol)
3.15 mg/mL (317 mL/g)

Ethyl Alcohol (Ethanol)
1.28 mg/mL (780 mL/g)
Isopropyl Alcohol (Isopropanol)
0.74 mg/mL (1350 mL/g)
