Yopo extraction. Failed ?

Here's the extraction steps:

1) Prepare citric acid solution by adding 1 gram of citric acid to 10 ml of acetone. We’ll use it later so put this aside, go onto step 2, but mix the citric acid solution occasionally. It will eventually all dissolve after about an hour or so.

2) Mix 100 grams of crushed Anadenanthera seeds (either peregrina or colubrina works) with and equal volume of pickling lime (calcium hydroxide) and add enough water to form a paste. Let it sit for about 1 hour, mixing it a few times. Dry it completely in an oven at 150 F.

3) Break apart the dried materal. Add 500 ml of acetone. Mix for about 5 minutes. Let the dry material settle to the bottom. The acetone should be reddish amber in color. Pour the acetone through a good quality filter.

4) Add 10 ml of citric acid solution (from step 1) to the filtered acetone and mix. Tiny crystals will form which look like yellow dust. Let the crystals settle to the bottom. Poor off the acetone, being careful not to pour off the crystals. Discard the acetone. Wash the crystals by adding 100 ml of fresh acetone, mix, let the crystals settle, and then carefully pour off the acetone and discard it (EDIT: you might want to let the acetone evaporate down to about 50 ml before discarding, as more precipitates might form. Keep in mind that a small amount of the alkaloids won’t precipitate out of the acetone. A small portion of this contains the tiny amount of DMT present in the seeds. If you save the acetone, you could extract the last bit of alkaloids using an A/B extraction, but It’s not really worth the trouble). Let all the acetone evaporate off the crystals. They’ll start absorbing water from the air and become dark brown goo once all the acetone has evaporated.

5) Mix the crystal goo with an equal volume of pickling lime. Add enough water to form a thin watery paste. Let it sit for about 1 hour while mixing a few times. Dry in an oven at 150 F.

6) Break apart the dried material. Dissolve in 100 ml acetone for about 5 minutes, mixing a few times. Filter off the pickling time. Evaporate the acetone leaving behind 5-HO-DMT freebase as a single solid mass of amber micro-crystals (and a few other active freebase alkaloids). It looks like amber candy, not crystals. It’s very difficult to form large crystals, so don’t bother. Just scrape it up and store in an air tight container away from light.

You can repeat steps 3-6 with the same seed mix up to 4 more times to recover nearly all the alkaloids if you like, but since the seeds are so cheap, it’s probably not worth the effort.

My friend had for unknown reason 40 g of Anadenanthera seeds. He decided to follow this tek. Everything seemed to work somehow well until step 6 (the yield was just very low, maybe 100 mg of goo).
But the second mix of lime+goo did not really dissolve in acetone. Once evaporated and filtrated, it leaves a half drop of greenish goo. My friend wonder why something that precipitated from acetone (acidified) do not want to dissolve again in acetone. He also wonder if he could try to smoke what was not dissolve...

The yield is too low. You should get at least 400 mg from 40 grams. The result should be amber brown material. It should NOT BE GREEN! Maybe you had the “fat seeds” which are bunk and produce green goo. Or maybe something was done wrong when following the tech? Try smoking one small hit and see if you get anything from it. One medium hit should produce definite visual effects. The “fat seeds” which produce the green goo, don’t have any alkaloids in them and aren’t really Anadenanthera.

SWIM had tried that tech many times with both real Anadenanthera peregrina and real Anadenanthera colubrina and it works just fine.

A. peregrina always produces more of a sticky result using that tech, but the result is amber brown, NOT GREEN. The tech works better with Anadenanthera colubrine producing more of a dry crystalline material. If it’s sticky you can usually cook out the sticky crap from A. peregrina by bringing the goo to 300 C. The sticky goo (likely DMT N-Oxide) will vaporize away leaving behind the freebase bufotenine which isn't sticky.

EDIT: SWIM later learned that all his "real" Anadenanthera peregrina was actually a variation of Anadenanthera colubrina being falsely sold as Anadenanthera peregrina!

The precipitate was a very dark-brownish goo and very sticky. The seeds were really looking what is seen on web pictures and the mix of lime-seeds smelt ammonia-like, so I guessed this was a good start.

Does the goo should completly dissolve in acetone, leaving only CACO3 powder ? Maybe it does require some heat ?

The goo should dissolve in the acetone.

CACO3 is completely insoluble in acetone. You can dissolve the goo completely in the acetone and filter out the lime or any CACO3 that forms. One thing that is VERY IMPORTANT is that after adding the water in all steps, make sure it's completely dry before going to the acetone steps otherwise your final product will have lots of contaminants.

According to SWIM, Yopo (Anadenanthera peregrina) is more toxic then Vilca (Anadenanthera colubrina), and produces more sticky results. If you heat the extract in an oven at about 500 F (260 C) for about 30 minutes, most of the sticky junk will vaporize away. The sticky stuff causes nausea. SWIM would like to know what the sticky stuff is exactly. It's NOT bufotenine. Bufotenine is not sticky. Freebase bufotenine vaporized at 608 F (320 C). After heating in the oven, most of the sticky stuff vaporizes away.

It should be active as is though. Have you tried it as is?

It might have plant oils in it. Does it smell or taste a little like peanuts? If so, there are plant oils in the goo preventing it from crystallizing. You may need to run the steps again on the goo to purify it more. Basically you dissolve the goo in acetone, and then add citric acid again to precipitate out the alkaloids once more (just as before). Then (just as before) you mix the precipitates with lime and a little water, and then when its dry you dissolve it in acetone again and dry the acetone. This should remove any oils and make the extract far more pure. If it’s still sticky, its from one of the alkaloids in the Yopo making it sticky. Some batches of Yopo produce more sticky results than other batches. Vilca usually produces dry results because it often mostly contains bufotenine which is dry, not sticky. DMT N-Oxide is sticky and oily. It can be vaporized away by heating the extract in an oven at about 500 F.

Actually when making Yopo or Vilca snuff, the seeds are traditionally roasted. This is said to reduce side effects. It also makes the extract less sticky if this is done before doing the tech on the seeds. SWIM has tried it both ways and finds the roasted seeds produce less nausea and a much less sticky extract. So, you can initially roast the seeds in an oven at about 500 F until they pop like popcorn before doing the tech. This should reduce the chance of getting sticky results. But again, this roasting at 500 F can also be performed on the goo after doing the extraction and the results are about the same.

Thanks for the details.
My friend did a mistake I guess. He evaporated the acidified acetone and recovered around 2.4 g of 'something' very sticky (including 1 g of citric acid) . So there was something in the acetone that did not precipitated. He mixed again that sticky thing with lime but there is now some acid citric in the sludge. Is there is a way to get rid of this acid ?

The lime will convert the citric acid into calcium citrate if enough is added. Calcium citrate is insoluble in acetone. Can he check the pH? If the pH is about 10-12 then he is fine. If it is lower then 10 he should add more lime. Make sure it is wet or it won't work. Add just enough water to make it like wet clay and mix it few a few minutes.

After mixing in the lime, let it dry completely. It must be bone dry. Once its bone dry he can extract with acetone again.

After extracting with acetone, then add citric acid to the acetone and then the alkaloids will precipitate out of the acetone again.

Carefully pour off the acetone leaving behind the alkaloids.

Mix the alkaloids with lime and enough water to make it wet but not watery. Then let it dry completely.

Now extract into acetone again.

Dry the acetone and you should have amber brown alkaloids that are very potent when vaporized. 10 mg should produce a very strong trip.

Did you friend ever test any of the results for activity?

Last steps are for purifying the freebase, but are there any other interrests to do them ? , I mean smoking with lime should not be a problem, lime will not vaporize with the light torch anyway.

Actually lime will help vaporize the alkaloids by preventing them from burning, and won't vaporize itself. At 512 C it decomposes into calcium oxide and water. If requires a temperature of 2850 C to vaporize calcium oxide.

DMT N-Oxide is sticky and won't crystallize and may be present in those seeds in large amounts sometimes. If so, the extract will be sticky unless DMT N-Oxide is somehow removed. It’s hard to remove it. You can heat the extract at 500 F in the oven to remove the DMT N-Oxide though. This causes it to vaporize away into the air.

Lime will also help keep the alkaloids from becoming sticky, so if there’s lots of DMT N-Oxide, leaving the lime in there might be a good thing.

Another option is to add calcium carbonate to the final extract if it is sticky. This works just like lime in keeping the extract dry. DMT N-Oxide is hygroscopic and absorbs water directly from the air. That’s why it’s sticky and gooey. Bufotenine is not at all hygroscopic or sticky.
